Title: A chemical perspective on the design of stable protein pharmaceuticals: polymers, particles, and immunogenicity

Abstract

The physical and chemical stability of proteins has a significant impact on the design of stable formulations, which can be administered to patients without adverse side effects such as, e.g., immunogenic reactions. In practice, the design of successful drug products is the result of close collaborations between scientists from multiple fields including biotechnology, protein and analytical chemistry, biophysics, pharmaceutical sciences and engineering. From a chemical point of view, some protein degradation pathways can be predicted, and depend on the chemical environment of the respective formulations. Other degradation processes frequently “come as a surprise”, and involve complex reactions of excipients, including polymers, or “left-over” material from manufacturing processes of chemicals and devices. This lecture will provide representative examples on the complexity of chemical degradation processes, and their potential consequences for formulation and device design.
